Tecnai F20 Problems

Instructions to Retrieve CCDBlankEnable

Film Jam

  • Access plate is jammed by lifting screen and looking through chamber viewing port.
  • Turn off pneumatics before attempting to dislodge or risk of losing finger

Computer Crash - what to do

  • Close column valves
  • Do not remove specimen
  • Reboot computer

V7 button

  • If a software problem prevents column valves closing; pressing Close V7 button (located on left side behind viewing screen) to close manually
    • Because of a bug, the TEM software is not happy if you press it an odd number of times, so after restart of computer, press it once more
    • If it was pressed an odd number of times, then HT and FEG will turn off upon opening valves. If this happens, press it once more, restart HT and try again.

Chiller failure - what to do

  • Make sure house chiller outside is operational. Check with Mike Goger, Dan Murphy or Kaushik Dutta.
  • Restart the F20's chiller in B-1
    • Remove the front panel and press reset button marked with an arrow.
    • Add ice to the water bath to start recirculation (temperature must be <70F to start)
    • Check insulated pipe to make sure cool water is going through the lines
  • Optionally, take this opportunity to reboot the TEM computer
  • You will need to restart:
    • The CCD Camera (physically turn it on)
    • The vacuum system
    • The HT
  • May need to restart TEM Server; probably best to simply reboot PC if this is the case

CCD camera restart

  • Press green button on camera box to start power
  • You will need to acquire new flafields in EMMENU
  • NOTE: PI-SCX box will not turn on until camera temperature reaches -17C

TEM Server restart

  • After a computer restart, TEM Server may not restart automatically (this seems to be fixed in new software as of March 2012)
  • Manually restart it as follows:
    • Go to Start menu > Programs > Administrative Tools > Services
    • Make sure TEM Server Status: started (right click and choose start if not)
  • Open the Tecnai User Interface
    • In High Tension panel move slider as high as software will allow (towards 200kV) to keep lenses warm

Restart the Vacuum and FEG

  • Make sure vacuum on
    • The banner on the Vacuum Expert Panel will give you the current status of the vacuum system.
    • If red, manually turn on vacuum - White Vac. button (do not trust if it is lit). You should hear valving and Vacuum (Expert) panel status should read "Starting up"
  • Make sure White HT button is lit. If not lit, press it.
  • Go to Setup > FEG control Panel
    • If "Operate" button is unlit in FEG Control turn on "Power"
    • Warm Start if under an hour, Cold Start if over an hour.
    • Choose Cold start if you do not know how long it was off. The warm or cold start determines how fast FEG reaches targets.
  • Select High Tension in the High Tension panel
    • Move High Tension to 200 kV, if not already. Check Measured HT: 201 kV
    • Make sure Gun lens: 3 (make sure by toggling between 2 and 3) and Extraction voltage: 3950. Click on arrow to ramp from 3400 to final target of 3950.
    • Make sure desired condenser aperture in

Login to data drive P:/

  • Data drive does not auto login. Manually remount. Password is on whiteboard

Necessary Alignments after restart of scope

Check detector configuration

  • Location: Camera > Detector Configuration.
  • Automatic mode off, Detector None. Current detector shift: On-Axis

Reload alignment files

  • Location: Tune > Alignment panel > File
  • Load most recent alignment file
  • Select all available alignments and move to "selected" side
  • Click "Apply"

Align scope

  • Center condenser aperture
  • In alignment panel follow Gun alignments
    • Gun Tilt - may be more than 1 bright spot, make sure not too close to aperture edge
    • Gun Tilt Pivot Points
    • Gun Shift - may have to do it several times
    • Check condenser aperture alignment again and condenser astigmatism
    • Spot size-dependent Gun Shift (optional) - spot 1 usually off
  • Do a final round of direct alignments
    • Beam tilt pp X and Y
    • Beam shift
    • Rotation center.

Condensor Stigmation

  • Spot sizes 1-5
  • Magnification 250kx
  • Condense beam
  • Press "stigmator" button
    • select "condensor"
  • Use "multifunction knobs" to minimize size of focussed beam

Check image of FEG Tip

  • Condense beam to point
    • Beam may not be round at crossover, this is the image of the FEG tip
    • 5mm circle = 10nm
    • You want the majority of your beam to fit within the 5mm circle

Check if beam is blanking properly

  • If not make sure CCD Blank Enable box present. CcdBlankEnable
    • Activating Tecnai Low Dose usually disables this box; must be re-enabled
  • Check pre-exposure time
    • Open "Pre-exposure" shortcut on Desktop
    • Under "Shutter Box Control" make sure pre-exposure time is 0 ms

Safe Microscope Shutdown

  • If you are told by NYSBC or FEI personel to shutdown the microscope
    • Press the Red Button on the right panel
      • This activates the "Standby Vacuum" which keeps IGP's on while shutting down the DP
      • Both red and green lights will remain on
